    目的:探索性建立阿司匹林抵抗(AR)的金黄地鼠动物模型,并进行评价。方法:选用健康雄性金黄地鼠130只,分为2组,造模组120只,普通饲料组10只。造模组采取高脂高糖高盐饲养12周,同时给予灌胃给药(0.74g/kg/d阿司匹林+2.22g/kg/d布洛芬)和吸烟4周,以建立阿司匹林抵抗(AR)模型。采用血小板聚集试验作为AR判定标准:腺苷二磷酸(ADP)诱导的血小板平均聚集率≥70%,花生四烯酸(AA)诱导的血小板平均聚集率≥20%,同时符合两项者为AR,符合其中一项者为阿司匹林半抵抗(ASR)。结果:多因素造模组,AR和ASR金黄地鼠的PAG均明显升高(P<0.01)。AR模型6只,发生率为5%;ASR模型28只,发生率为23%。结论:通过对AR产生的病理环境的模拟,多种危险因素的综合作用,实验成功制造出AR金黄地鼠动物模型。

    Abstract:

    Objective:To establish aspirin resistance(AR) model in hamster and evaluation.Methods:130 male hamsters,were selected and divided into model group(120) and control group(10).High fat,sugar and salt were fed for 12 weeks,meanwhile oral administration of 0.74 g/kg/d aspirin +2.22 g/kg/d ibuprofen and smoking were given for 4 weeks to build the Aspirin resistance(AR) model.Platelet aggregation(PAG) test was used as the criteria of AR:if adenosine diphosphate(ADP) induced average PAG rate≥70%,and arachidonic acid(AA) induced average PAG rate≥20%,it was AR;if only one item met the criterion,it was aspirin semi resistance (ASR).Results:The PAG was significantly increased(P<0.01) in muliti-factor model,AR and ASR hamster.AR model built in 6 animals,the rate was 5%;ASR model built in 28,the rate was 23%.Conclusion:The pathological environment simulation,combined with the effect of multiple risk factors,experimental AR models are successfully established in hamsters.
